His research focused on immune response to cancer and metastases, and infection. In 2013 he was investigated for breaches of research integrity by the University of Melbourne following reports from his colleagues at the Peter MacCallum Cancer Centre. He was cleared on weak evidence. From 2013-2021 Smyth headed the Immunology in Cancer and Infection Laboratory at the QIMR Berghofer Medical Research Institute, Brisbane.
