Her research was the first to the show a HIV protein functioning as an ion channel. She returned to the University of Alabama at Birmingham (UAB) as a postdoctoral fellow in the Center for AIDS Research from 1998 to 2000. Piller then came back to Australia in 2000 when she received a Young Investigator Award from the Centre for Immunology (CFI), the research campus of St Vincent's Hospital, Sydney. Her work as a senior research officer and group head involved her in various projects looking at Vpr and gp41. Piller also lectured in the Department of Medicine at the University of New South Wales and became a visiting fellow in the Division of Biochemistry and Molecular Biology at the John Curtin School of Medical Research (JCSMR).
