Cooke remained loyal to the University of Adelaide and returned there to lecture after completing studies in England. He spent 43 years teaching at the University and was eventually appointed Reader in Chemistry. Cooke was a foundation member of the Australian Chemical Institute and served the South Australian Branch in executive positions for at least twenty years.
