His research focused on dispersal strategies of parasites, leading him to propose that the need for an intermediate host was a primitive characteristic, infection of hosts by ingestion of the parasite's embryonated eggs being a more advanced strategy. He described 10 genera and 23 species new to science. Sprent was Professor of Parasitology at the University of Queensland from 1956 to 1983 where he established the Department of Parasitology. In 1987 and 2005 Sprent donated to the Queensland Museum his collections of 7,600 nematode specimens, 30,000 microscope slides, correspondence and a substantial number of scientific reprints.
