Charles MacKay graduated in medicine at the University of Melbourne in 1905. He then worked at Melbourne Hospital, the Children's Hospital and the Infectious Diseases Hospital. Upon the outbreak of World War I, he joined the Royal Army Medical Corps in England. As lieutenant-colonel, he later headed the General Hospital in Saloniki, where he became a specialist in rehabilitation and rheumatology. On returning to Australia in 1936, he was medical assistant to the director of the Australian Institute of Anatomy, Canberra, in 1936, and Acting Director in 1937. From 1939-1942, he was the director of the Anti-Cancer Council of Victoria. From 1942-1945, he was wartime executive medical officer of the Medical Equipment Control Committee, and then joined the Cancer Institute as a secretary and later as executive medical officer.
