It commenced formal operations early in 1908. The Department administers policy and Acts of Parliament relating to rural industries including scientific research, industry education and extension.

In 2004 the Department of Agriculture merged with other New South Wales government agencies to form the Department of Primary Industries. The other agencies were the Department of Mineral Resources, NSW Fisheries, the Forestry Commission, and State Forests. In 2009 the Department of Primary industries was amalgamated into the Department of Industry and Investment, and now exists as a department in the Department of Regional NSW.
